It is entirely possible to set up a payroll yourself, although you do need to be reasonably comfortable using a computer. If so you can either buy an off-the-shelf payroll package or use the HM Revenue & Customs (HMRC) online payroll scheme.
But first, to get started HMRC has a useful guide – PAYE for employers: the basics at
http://www.hmrc.gov.uk/paye/intro/basics.htm
This tells you all you need to know to get started as an Employer. If you have any queries you can call HMRC Employer Helpline on 08457 143 143. It is open from 8.00am to 8.00pm Monday to Friday and 8.00am to 4.00pm on Saturdays
To register for the HMRC online payroll scheme, you will need to quote your Employer PAYE reference and your Accounts Office reference.
All the information you need to start up the HMRC payroll scheme is available at: http://www.hmrc.gov.uk/paye/file-or-pay/fileonline/register.htm
The Online Service Helpdesk can be reached on 0845 60 55 999. It is open the same hours as Employer Helpline.
HMRC also runs workshops and presentations to help new employers with online filing. The HMRC website has details.
As an alternative to using HMRC you might want to consider using a payroll bureau or an accountant. They will take care of your payroll processing. There are a number of payroll bureau so costs are very competitive. The main thing to bear in mind is that payroll bureau have many customers and work to strict deadlines to meet processing requirements. Many accountants offer payroll service generally at a higher cost than payroll bureau but sometimes on a more accommodative basis to their customer.
