Q: I want my employees to start taking their holidays in blocks of five days rather than one day at a time. Can I do this?

In general, yes. You are entitled to refuse requests for leave or to specify when leave can be taken. So you can inform your employees that you will only approve holiday that has been properly requested and if it is in blocks of five days.
However, it may be worth considering allowing some of the days to be taken individually to allow for specific events, such as appointments, weddings, deliveries, utilities visits, and emergency repairs. You should also consider that you may need to consider an amount of flexibility to recognise the need for reasonable adjustments for disabled people, those with dependents and anyone who may need the time off for religious festivals. Also consider that if people book leave that incorporates any public or bank holidays, they may not use up five days holiday to have five days off work, so you will need to work out how you will deal with the odd days.
